I think she is knitting a sock. There is a ball of yarn on the floor. The black splot is probably dropped ink, and she has what appears to be a line smear that makes it look like she has a mustache…

I think the gag here is about women not understanding when something is funny instead of serious, and I tend to agree; my mother is the only person I know who was depressed by Who Framed Roger Rabbit, because she thought it was meant to make fun of the mentally ill. Seriously.
